Methanesulfonyl chloride (232 1iL, 3.00 mmol) was slowly added at OC to a solution of tert-butyl (28)-2-methylpiperazine- 1 -carboxylate (300 mg, 1.5 mmol) and triethylamine (835 1iL, 6 mmol) in methylene chloride (6 mL). After stirring at r.t. for 1 h, reaction mixture was carefully quenched by addition of water and the desired product was extracted with ethyl acetate. The organic phase was washed with brine, dried over sodium sulfate, and filtered.The filtrate was concentrated under reduced pressure. The resulting residue was purified by Biotage Isolera (flash purification system with hexane/ethyl acetate at a ratio from 0 to 100%) to give tert-butyl (25)-2-methyl-4-(methylsulfonyl)piperazine- 1 -carboxylate (404 mg, 97%).
