129722-25-4, To a solution of dehydro-Aripiprazole (1.5 g, 3.36 mmol)2-methyltetrahydroffiran (30 mE) was added silver carbonate (1.853 g, 6.72 mmol) and hexyl iodomethyl carbonate (2.021 g, 7.05 mmol) in 2-methyltetrahydrofuran (4 mE) at room temperature. The reaction was stirred for 4.5 days. The reaction was quenched with H20 (30 mE) and filtered through celite. The reaction was extracted with ethyl acetate (3×20 mE), washed with brine (20 mE), dried over MgSO4 and concentrated. To a mixture of dehydro-aripiprazole (1.5 g, 3.4 mmol), potassium tert-butoxide (0.75 g, 6.7 mmol) and 2-methyltetrahydrofuran (30 mL) at 0 C. was added hexyl chloroformate (1.32 mL, 8.1 mmol). The reaction mixture was stirred for 2 h, allowed to self warm to room temperature and stirred for a further 4 h. The reaction mixture was then diluted with water and extracted with ethyl acetate. The organic phase was dried over MgSO4, filtered and evaporated. The residue was further purified on silica eluting with ethyl acetate/tetrahydrofuran and the major product containing fractions evaporated to give a yellow solid. This was triturated in heptane (30 mL) for 2.5 h, filtered and dried to give Compound 328 (1.55 g) as a white solid.1H-NMR (300 MHz, CDCl3) delta 8.15 (d, 1H), 7.71 (d, 1H), 7.32 (d, 1H), 7.21-7.12 (m, 3H), 7.09 (d, 1H), 6.97-6.91 (m, 1H), 4.29 (t, 1H), 4.13 (t, 1H), 3.12-3.01 (m, 4H), 2.73-2.55 (m, 4H), 2.50 (t, 2H), 1.95-1.85 (m, 2H), 1.82-1.70 (m, 4H), 1.50-1.28 (m, 6H), 0.94-0.86 (m, 3H). General procedure: To a solution of dehydro-Aripiprazole (1.0 g, 2.24 mmol) in 2-methyltetrahydroffiran (25 mE) was added silver carbonate (0.864 g, 3.13 mmol) and iodomethyl octanoate (0.764 g, 2.68 mmol) at room temperature. The reaction was stirred for5 days. The reaction was quenched with H20 (30 mE) and filtered through celite. The reaction was extracted with ethyl acetate (3×20 mE), washed with 5% w/v sodium sulfite solution (15 mE), brine (20 mE), dried over MgSO4 and concentrated. A mixture of (S)-l-[((9H-Fluoren-9-yl)methoxy)carbonyl)-4-(tert- butoxycarbonyl)piperazine-2-carboxylic acid (0.27 g, 1.0 mmol), tert-butyl [2- (aminomethyl)-4-chlorobenzyl]carbamate (0.5 g, 1.10 mmol), HOBT (0.135 g, 1 mmol), and EDOHC1 (0.25 g, 1.30 mmol) in anhydrous DMF (3 mL) was stirred under nitrogen at room temperature for 16 h. The reaction mixture was quenched with water (10 mL), diluted with EtOAc (25 mL) and washed sequentially with saturated aqueous aHC03 solution (25 mL) and brine solution (25 mL). The organic layer was separated, dried over anhydrous Na2S04, filtered and concentrated. The residue was purified by reverse phase combiflash column chromatography (CI 8; eluent: 10-100percent acetonitrile: water) to afford the title compound (0.61 g) as white solid.
