AIE-based nanoaggregate tracker: high-fidelity visualization of lysosomal movement and drug-escaping processes

High-fidelity imaging and long-term visualization of lysosomes are crucial for their functional evaluation, related disease detection and active drug screening. However, commercial aggregation-caused quenching probes are not conducive to precise lysosomal imaging because of their inherent drawbacks, like easy diffusion, short emission and small Stokes shift, let alone their long-term tracing due to rapid photobleaching. Herein we report a novel aggregation-induced emission (AIE)-based TCM-PI nanoaggregate tracker for direct visualization of lysosomes based on the building block of tricyano-methylene-pyridine (TCM), wherein introduced piperazine (PI) groups behave as targeting units to lysosomes upon protonation, and the self-assembled nanostructure contributes to fast endocytosis for enhanced targeting ability as well as extended retention time for long-term imaging. The piperazine-stabilized TCM-PI nanoaggregate shifts the emission maximum to 677 nm in an aqueous environment, and falls within the desirable NIR region with a large Stokes shift of 162 nm, thereby greatly reducing biological fluorescent background interference. In contrast with the commercially available LysoTracker Red, the essential AIE characteristic of high photostability can guarantee three-dimensional high-fidelity tracing with low photobleaching, and little diffusion from lysosomes, and especially overcome the AIE bottleneck to target specificity. Consequently, the AIE-based nanoaggregate tracker successfully achieves the high-fidelity and long-term tracing of lysosomal movement and even monitors the drug-escaping process from lysosomes to cell nuclei, which provides a potential tool to benefit drug screening.

Synthesis, molecular docking studies, and in vitro antimicrobial evaluation of piperazine and triazolo-pyrazine derivatives

For this work, two series of new piperazine derivatives (3a-o) and triazolo-pyrazine derivatives (3p-t) were synthesized in a single-step reaction. All twenty adducts were obtained in good to high yields and fully characterized by H-1 NMR, C-13 NMR, IR, and mass spectrometry techniques. To further confirm the chemical identity of the adducts, a crystal of N-{[(4-chlorophenyl)-3-(trifluoromethyl)]-5,6-dihydro-[1,2,4]triazolo[4,3-a]}pyrazine-7(8H)-carboxamide (3t) was prepared and analyzed using X-ray crystallography. In vitro screening of the antimicrobial activity of all compounds (3a-t) was evaluated against five bacterial and two fungal strains. This study disclosed that N-{[(3-chlorophenyl)]-4-(dibenzo[b,f][1,4]thiazepin-11-yl)}piperazine-1-carboxamide (3o) was the superior antimicrobial with good growth inhibition against A. baumannii. Furthermore, the results from the performed molecular docking studies were promising, since the observed data could be used to develop more potent antimicrobials.

Synthesis and Bioactivities of Novel Pyrazole Oxime Derivatives Containing a N-Pyridylpyrazole Moiety

In search of novel pyrazole oxime compounds with potent bioactivities, twenty pyrazole oxime derivatives were synthesized by introducing N-pyridylpymzole unit into pyrazole oxime skeleton, based on the lead of fenpyroximate. The title compounds were structurually characterized by H-1 NMR, C-13 NMR and elemental analysis. The preliminary bioassay showed that all the title compounds had 100% insecticidal activities against Mythimna separata Walker at 500 mu g/mL. Five compounds displayed 100% mortality rate towards Mythimna separata Walker at 100 mu g/mL. Two compounds both exhibited 40% mortality rate against Mythimna separata Walker at 20 mu g/mL. In addition, three compounds had 40 similar to 60% insecticidal activities to Aphis medicaginis at 500 mu g/mL. One-pot, two-step synthesis of 7-methylene-1,5-piperazine-fused 1,2,3-triazoles

A facile, one-pot two-step synthesis of 7-methylene-1,5-piperazine-fused 1,2,3-triazole derivatives has been developed. The protocol employs an N-allylation of N-propargylated amines with 2,3-dibromopropene in the presence of K2CO3 in DMSO and a CuI-catalyzed [3 + 2] cycloaddition reaction of the synthetic N-(2-bromoallyl)-N-propargyl amines with sodium azide sequentially. Such a method provides methylene-substituted 1,2,3-triazole fused piperazines with some advantages such as simple operation, high efficiency and good product yield (80-91%) through readily available starting materials.

Bioreducible cationic random copolymer for gene delivery

Cationic polymers have been widely investigated for gene delivery, although their low transfection efficiency and high cytotoxicity limit their application. We synthesized a bioreducible cationic random copolymer, poly(cystamine bisacylamide-aminoethyl piperazine)-co-poly(cystamine bisacylamide-histamine) (denoted as CBA-AEP-His) fromN,N ‘-cystamine bis acrylamide (CBA) with aminoethyl piperazine (AEP) and histamine (His). CBA-AEP-His copolymer possesses disulfide linkages that endow it with redox-responsivity to the intracellular environment. This polymer efficiently condenses pZNF580 into complexes with the size of 160 +/- 4 nm to 280 +/- 5 nm and positive zeta potential of 20 +/- 0.3 mV to 30 +/- 0.4 mV. The gel-retardation assay shows that CBA-AEP-His can retard pZNF580 even at a low mass ratio of 1/1. The gene complexes were triggered to release pZNF580 when exposed to the reducing environment of dithiothreitol (DTT). CBA-AEP-His random copolymer presented higher buffer capacity owing to its His moieties, which protected pZNF580 from DNase degradation. The gene transfection results reveal that CBA-AEP-His can efficiently deliver pZNF580 and transfect EA. Hy926 cells. The MTT assay indicates that CBA-AEP-His and its complexes exhibit lower cytotoxicity than PEI25KDa. These results illustrate that CBA-AEP-His had promising properties for gene delivery, which may provide a suitable platform for the development of a non-viral gene carrier.

Fabrication of new magnetite based sulfonic-phosphotungstic dual-acid catalyst for catalytic acetalization of benzaldehyde with ethylene glycol

A new sulfonic-phosphotungstic dual-acid hybrid catalyst based on silica coated magnetite nanoparticles (SCMNPs) containing two types of Bronsted acidic sites i.e. sulfonic acid (-SO3H) and phosphotungstic acid (HPW) groups, was prepared with chemical and electrostatic interactions of these acidic functional moieties with piperazine-grafted propylsilyl spacer groups. Physicochemical techniques including FT-IR spectroscopy, elemental analysis and inductively coupled plasma-optical emission spectroscopy (ICP-OES), vibrating sample magnetometry (VSM), X-ray diffraction (XRD), energy dispersive X-ray (EDX) analysis, scanning electron microscopy (SEM), and transmission electron microscopy (TEM) were used to characterize the prepared solid acid catalyst. Acetalization reaction between benzaldehyde and ethylene glycol was done to investigate the catalytic activity of prepared catalyst and 97% conversion toward acetal production was reached in short reaction time. The lower conversion was also achieved for the same catalyst without any HPW species, demonstrated the positive role of the second acidic sites on progression of this reaction. In addition, this easily separable solid acid catalyst reused for four runs with no observable loss in benzaldehyde conversion.

Synthesis and biological evaluation of ethacrynic acid derivatives bearing sulfonamides as potent anti-cancer agents

A series of ethacrynic acid (2-[2,3-dichloro-4-(2- methylidenebutanoyephenoxy]acetic acid) (EA, Edecrin) containing sulfonamides linked via three types of linkers namely 1,2-ethylenediamine, piperazine and 4-aminopiperidine was synthesized and subsequently evaluated in vitro against HL60 and HCT116 cancer cell lines. All the EA analogs, excluding 6a and 6c, showed anti-proliferative activity with IC50, in the micromolar range (less than 4 uM). Three derivatives 6b, 7 b and 7 e were selected for their interesting dual activity on HL60 cell line in order to be further evaluated against a panel of cancer cell lines (HCT116, A549, MCF7, PC3, U87-MG and SKOV3) as well as on MRCS as a normal cell line. These compounds displayed IC50 values in nanomolar range against A549, MCF7, PC3 and HCT116 cell lines, deducing the discovery that piperazine or 4-aminopiperidine is the linker’s best choice to develop EA analogs with highly potent anti-proliferative activities own up to 24 nM. Graphene oxide interlayered thin-film nanocomposite hollow fiber nanofiltration membranes with enhanced aqueous electrolyte separation performance

In this study, a kind of thin-film nanocomposite (TFN) hollow fiber nanofiltration (NF) membranes was fabricated via incorporating graphene oxide (GO) interlayer using the interfacial polymerization (IP) reaction between piperazine (PIP) and trimesoyl chloride (TMC). The surface morphology and cross-sectional structure of the fabricated TFN hollow fiber NF membrane were extensively investigated. The effects of the PIP concentration, species of the aqueous additives, and the GO loading on the structure and performance of the hollow fiber NF membrane were also investigated in detail and the optimal preparation conditions were achieved. Fourier transform infrared spectroscopy (FTIR) confirmed the successful introduction of the GO interlayer into the composite hollow fiber NF membrane. Moreover, the incorporation of GO interlayer helped to reduce the skin thickness of the composite hollow fiber NF membrane remarkably, and thus helped to increase greatly the water permeance while maintaining a high salt rejection. Under an optimal GO loading of 20.0 mg m(-2), the TFN hollow fiber NF membrane achieved a water permeance of 80 L m(-2) h(-1) MPa-1 and a Na2SO4 rejection of 96.1% for 2000 mg L-1 aqueous Na2SO4 solution, much higher than the interlayer-free thin-film composite (TFC) membrane. Meanwhile, the TFN hollow fiber NF membrane showed an excellent selectivity of Na2SO4 over NaCl, as well as good fouling resistance and long-term stability, demonstrating the vast potential in application for the selective separation of monovalent salt and divalent salt.

Enhanced research of absorption by mass transfer promoters

Activators such as piperazine (PZ) are widely used to increase the CO2 absorption rate of aqueous methyl-diethanolamine (MDEA) solutions by accelerating reaction. However, a big difference (1 to 2 orders of magnitude) in the enhancement of absorption rate with the addition of PZ was obtained in different studies. In this work, it was proved that mass transfer is the rate-limiting step in the above process and in most cases enhancing mass transfer is quite essential. Some low-polar solvents were screened as mass transfer promoters (MTPs) to enhance mass transfer by inducing interfacial turbulence, which markedly improved the CO2 absorption performance of MDEA solutions. The individual addition of PZ and MTPs to the MDEA solution, which correspond to the reaction enhancement and the mass transfer enhancement, increased the absorption rate by 131% and 33%-319%, respectively. The co-effects of enhancing both reaction and mass transfer (in the presence of activators and MTPs) can lead to 2 to 10-fold increase in the absorption rate. Furthermore, MTPs could increase the CO2 cyclic loading and desorption rate by 27% and 100%, respectively, indicating that the MDEA/MTPs had significant advantages for reducing the energy consumption in CO2 capture applications.

Application of diazonium-induced anchoring process on ultrafiltration substrate for the fabrication of nanofiltration membrane with enhanced desalination performance

In membrane process, the lateral transport path of water molecule through substrate has a great impact on the permeance of thin-film composite (TFC) membrane, thus the optimization of substrate by constructing interlayer without affecting its pore structure is desired. Here, we report a facile, scalable and universal surface grafting strategy based on a diazonium-induced anchoring process (DIAP), for which an ultra-thin polyaminophenylene (PAP)-like layer is covalently grafted on the substrate. The PAP-like layer significantly increases the wettability of surface and pore as well as surface positive charges of substrate, but has little infiuence on its surface pore size and structure. Moreover, the effects of the anchored PAP-like layer on the distribution, release and uptake of piperazine (PIP) monomers during interfacial polymerization (IP) process are investigated systematically. The polyamide (PA) nanofiltration (NF) membrane made on this substrate features a highly cross-linked, thinner and smoother PA layer with less PA invasive, and highlights a 2-fold water fiux increase and simultaneously enhanced divalent salts (Na2SO4, MgSO4 and MgCl2) rejections (above 98.0%). Thus, combined with the superior structural stability and antifouling property, the obtained TFC membrane holds great potential in water soft ening and household water purification.
