The mixture was stirred for 16 h at 90 °C, diluted with water and extracted into DCM (20 mL). The organic layers were passed through a phase separator and concentrated and the crude mixture was purified by flash silica column chromatography. Following method I from compound 15c (250 mg, 0.76 mmol) and /so-propylpiperazine (125 muIota, 0.86 mmol). Purification using flash silica column chromatography (gradient elution /’-hex/EtOAc 0percent to 100percent) gave the title compound as a yellow oil (187 mg, 65percent). LCMS (ES+) 379 (M+H)+

The resultant was stirred for 1 hour, and then cooled to 0C. Sodium triacetoxyborohydride (31.6 g, 150 mmol) was added in portions. The resultant was reacted at room temperature for 6 hours. The solvent was evaporated, and the residue was dissolved by adding ethyl acetate. The resultant was washed by water, dried, concentrated, and purified by silica gel column chromatography to give (S)-1-benzyl-2-methyl-4-(1-methylpiperidin-4-yl)piperazine (7.86 g, 76% yield). MS m/z [ESI]: 288.2 [M+1].

5308-25-8, A mixture of 7-chloro-1-cyclopropyl-6-fluoro-4-oxo-1,4-dihydroquinoline-3-carboxylic acid 1a (1 mmol) and Nethylpiperazine2y (1.5 mmol) and n-FZSA (0.06 g) as catalystin H2O (5 ml) were heated under reflux for the appropriatetime. The reaction was monitored by TLC. After appropriatetime, the catalyst was separated using an externalmagnet and washed with hot ethanol (5 mL). The reactionmixture was then cooled to room temperature. The precipitatedsolid was collected by filtration, and recrystallized fromethanol 96% to give desired compound in high yields.

The solvent was removed under high vacuum. The residue was taken up in CH2C12 and washed with an aqueous saturated solution of NaHCO3. The organic layer was separated and dried over Na2504 and concentrated in vacuo. The crude was purified by column chromatography (5i02, CH2C12/MeOH=95/5 to 90/10) to afford the title product (40 mg, 63%) as a light yellow solid.

A solution of 2,4-dichloro-5,7-difluoro-3-nitroquinoline (5.58 gm, 2.0×10?2 moles) in 2-methyl tetrahydrofuran (50 mL) is stirred as diisopropylethylamine (2.84 gm, 2.2×10?2 moles) and N-2-aminoethyl-N? methylpiperazine (3.15 gm, 2.2×10?2 moles) are added. This solution is stirred at room temperature overnight. The yellow reaction mixture is diluted with more 2-methyl-tetrahydrofuran (50 mL) and this is washed with water (100 mL) followed by brine (50 mL). After being dried over magnesium sulfate, the solution is filtered and the solvent is removed under reduced pressure. The oily residue is stirred with diethyl ether (25 mL) and this is cooled on ice causing the product to crystallize. The solid yellow product is isolated by filtration, washed with ether and dried. The yield is about 4.32 gm.

3.49 g (17.0 MMOL) 4-CHLOROBENZYLBROMIDE was added dropwise to a mixture of 2.04 G (20.4 MMOL) 2-Methyl piperazine and 2.40 ml (17 MMOL) triethylamine in 60 ml DMF at room temperature. The reaction mixture was stirred for 16 hours at room temperature, the poured onto aq. sodium bicarbonate solution and extracted with ethylacetate. Purification of the crude product by flash chromatography gave 2.26 g (10.1 mmol, 59 %) of 1- (4-chloro- benzyl)-3-methyl-piperazine as colorless oil. 1H-NMR (400 MHz, DMSO-d6): 0.91 (d, 3H), 1.56 (t, 1H), 1.90 (TD, 1H), 1.91-2. 03 (m, 1 H), 2.56-2. 74, m, 4H), 2.79 (dt, 1 H), 3.40 and 3.44 (AB-Sys., 2H), 7.33 (d, 2H), 7.40 (d, 2H).
