To a solution OF RAC-3-METHYL-PIPERAZINE-L-CARBOXYLIC acid tert-butyl ester (350 mg, 1.75 mmol) in dimethylformamide (3 ml) were added 2-Iodo-5-methanesulfonyl-benzoic acid (540 mg, 1.67 mmol), N-ETHYLDIISOPROPYLAMINE (1. 8 ml, 10.5 mmol), and TBTU (630 mg, 1.92 mmol). The reaction mixture was then allowed to stir at room temperature for 16 hours. The reaction mixture was then concentrated in vacuo and the residue was purified by column chromatography (SI02, 20g, Heptane/EtOAc 0-100percent) to give the title compound (400 mg, 45 percent). MS (m/e): 526.2 (M+NH4+).

68104-63-2, To a solution of 6.1 mmol 2-iodo-5-methanesulfonyl-benzoic acid in 20 ml dimethyl formamide were added successively 6.75 mmol TBTU, 43 mmol N-ethyldiisopropylamine and 6.75 mmol 4-piperazin-1-yl-benzonitrile (commercially available, e.g. from Fluka). The reaction mixture was stirred at 20 C. for 1.5 h and then concentrated in vacuo. Addition of 200 ml water followed by filtration yielded the crude product which was recrystallized from methanol to afford the title compound as a white solid (yield 87%). MS (m/e): 495.9 (M+H+, 100%).

The DMF was then removed under reduced pressure and the residue partitioned between water and methylene chloride. The methylene chloride was drawn off, dried and evaporated to a solid to afford 42-2. This recovered product was treated with neat trifluoroacetic acid and the excess trifluoroacetic acid evaporated off. The resulting residue was partitioned between methylene chloride and aqueous Na2CO3. The free base did not extract from the aqueous so the aqueous was evaporated to dryness, washed the methanol, and the solids filtered off to afford 42-2. 1H-NMR(CD3O): 3.69(t, 4H); 3.59 (t, 2H); 3.54(t, 2H); 3.22(s, 2H); 2.83(t, 2H); 2.77(t, 2H); 2.49(t, 4H).

5747-48-8, A 1 liter round bottom flask equipped with stirring rod, thermo pocket, water condenser was charged with solution of 11- piperazinyldibenzo [b,f] [1,4] thiazepine in toluene 350 cc [63.0 g (0.22 moles)] and the mixture was stirred for 15 min at 25- 30C. Sodium carbonate [41.0 gm (0.39 moles)], tetra butyl ammonium bromide [16.0 gm (0.05 mole)] and 2-(2-chloroethoxy)ethanol [32.0 gm (0.257 moles)] were added at room temperature. The reaction mixture was heated to reflux at 110-112C. The reaction mixture was maintained at reflux for 10-12 hrs. The reaction mixture was analyzed by HPLC (to check the absence of compound of Formula IV) and was cooled to 25C to 30C, and was added 150 cc DM water. The reaction mixture was then stirred for 30 min at 25-30C. The layers were separated and the aqueous layer extracted with 50 cc toluene. The extract and the organic layer were combined, to which was added 250 cc water and was acidified with formic acid to obtain a pH of 2- 3. The reaction mixture was stirred for 30 min at 25-30C. The layers were separated and the aqueous layer washed with 100 cc toluene twice. To the aqueous layer was added 250 cc toluene, and the pH adjusted to 8-10 using sodium carbonate. The resulting reaction mixture was stirred for 30 min at 25-30C. The layers were separated and the aqueous layer extracted with 125 cc toluene. The organic layers were combined and washed with DM water 300 cc twice. The organic layer was distilled off under vacuum below 70C to afford 2-(2-(4-dibenzo[b,fJ-[l,4] thiazepine- l l-yl-l-piperazinyl)ethoxy) ethanol. Purity of 2-(2-(4-dibenzo[b,f]-[l,4] thiazepine- l l-yl-l-piperazinyl)ethoxy) ethanol was 99.0% (area % by HPLC).
