The resulting mixture was stirred at room temperature for 1.5 h then partitioned between AcOEt and water. The layers were separated and the aqueous phase was extracted three times with AcOEt. The combined organic phases were washed three times with brine, dried over MgS04 and concentrated in vacuo. Purification of the residue by SP4 using a 25 G silica cartridge (gradient: 13 to 63% AcOEt in Hexanes) gave 1 ,1 -dimethylethyl 4-[(4-bromophenyl)methyl]-3-oxo-1 – piperazinecarboxylate (763 mg, 2.066 mmol, 83 % yield) as an oil which solidified to a white solid over 16 h. A solution of 6-chloro-1-(3,5-diisopropylpyridazin-4-yl)-7-(2-fluorophenyl)pyrido[2,3-d]pyrimidine-2,4(1H,3H)-dione (0.439 g, 0.967 mmol, Intermediate 212), phosphoryl trichloride (0.108 mL, 1.16 mmol), and DIPEA (0.168 mL, 0.967 mmol) in acetonitrile (5 mL) was stirred under an air condensor and drying tube at 80 C. for 30 min. The reaction mixture was cooled to RT, and tert-butyl (2S,5R)-2,5-dimethylpiperazine-1-carboxylate (0.207 g, 0.967 mmol; eNovation Chemicals LLC, Bridgewater, N.J.) was added; the solution was stirred a RT for 30 min. The reaction mixture was diluted with EtOAc (200 mL), added to a separatory funnel, and washed with saturated, aqueous sodium bicarbonate (2*100 mL); the organic layer was separated, dried over anhydrous Na2SO4, and concentrated in vacuo. The reaction mixture was diluted in an aqueous saturated NaHC03 solution and filtered. Preparation 33: (R)-4-(5-Carboxy-pyridin-2-yl)-3-methyl-piperazine-1-carboxylic acid tert-butyl ester A mixture of 6-fluoronicotinic acid (150 g, 1.063 mol) and (R)-3-methylpiperazine-1-carboxylic acid tert-butyl ester (234.2 g, 1.169 mol) in tetrahydrofuran (1.75 L) was cooled to -40 C and then 2 M isopropylmagnesium chloride in tetrahydrofuran (1.196 L, 2.39 mol) was added slowly maintaining the temperature less than -20 C. The reaction mixture was slowly warmed to RT, stirred at RT for 4 h and then 1 N HCl (1.75 L) and water (1.175 L) were added. The reaction mixture was extracted with ethyl acetate (4 L). The organic phase was evaporated to provide crude solid (534 g). To the crude solid was added acetone (2 L) and water (200 mL). The resulting reaction mixture was heated to 50 C and then water (2.8 L) was added slowly. Seed crystals from a previous run at smaller scale were added after ~ 1 L of water. The reaction mixture was cooled to 20 C over 3 h, stirred at 20 C overnight and filtered. The solid was washed with 2:3 acetone:water (2 x 500 mL) and dried under vacuum to provide the title compound (329 g, 96 % yield) as an off-white solid. A 50 mL rb flask equipped with an air condenser, rubber septum and a magnetic stir bar was set under Ar atmosphere and charged with 500 mg of 1 -(/tv7-b utoxy carbonyl)piperazine-2- carboxylic acid (2.17 mmol, 1 equiv.), 80.0 mg of DMAP (0.651 mmol, 0.3 equiv.), 500 mg of EDCI (2.61 mmol, 1.2 equiv.), 10 mL of CH2CI2 and 3 mL of dry methanol. The reaction mixture was stirred at 40 C for 1.5 h and then the reaction was allowed to cool to rt. After stirring for 20 h, the reaction mixture was concentrated and the residue was dissolved in CH2CI2. The CH2CI2 solution was washed with water (3x). The water solutions were combined and the product was back-extracted with CH2CI2 (3x). Then the combined organics were washed with sat. NaHC03 solution, brine and dried over Na2S04. The crude product (661 mg) was purified on silica gel column using 0 to 5 % MeOH in CH2CI2 as eluent affording 419 mg (79 %) of the product 41 as a colorless oil.

780753-89-1, Example 13 4-[(2,3-Dichlorophenyl)carbonyl]-1 -(4-fluorophenyl)-2-piperazinone (E13); 1-(4-Fluorophenyl)-2-piperazinone (150mg, 0.77 mmol, prepared as described above for Example 12) and lambda/-ethyl-lambda/-(1-methylethyl)-2-propanamine (0.20 ml, 1.16 mmol) were added together in dichloromethane (4 ml) at O0C. Subsequently 2,3- dichlorobenzoyl chloride (178 mg, 0.85 mmol) was added portionwise. The mixture was left under argon and in an icebath and allowed to return to room temperature whilst being stirred constantly overnight. Dichloromethane and aqueous 3N citric acid were then added and the mixture was extracted into dichloromethane (x2). The dichloromethane layers were combined and washed sequentially with water (x1 ), saturated aqueous sodium hydrogen carbonate (x1 ), water (x1 ), and brine (x1 ), and then dried over magnesium sulphate. The solvent was evaporated in vacuo to give 4-[(2,3-dichlorophenyl)carbonyl]-1-(4-fluorophenyl)-2-piperazinone (224 mg) as a white solid. LC/MS [M+H]+ = 367, retention time = 2.62 minutes.

The reaction mixture was added with water and was washed with hexane/ethyl acetate (4/1) to remove impurities. After removing impurities, the reaction mixture was extracted with ethyl acetate, and the organic layer was washed with water and saturated brine and dried over anhydrous magnesium sulfate. Then, the solvent was evaporated under reduced pressure and the residue was purified by silica gel column chromatography (ethyl acetate) to obtain 4-[4-(2-methoxyethyl)piperazin-1-yl]-2-nitrobenzaldehyde (0.23 g, 68%). 1H-NMR (300 MHz, CDCl3) delta 2.63-2.69 (m, 8H), 3.38 (s, 3H), 3.49 (t, J = 5.1 Hz, 2H), 3.56 (t, J = 5.3 Hz, 2H), 7.04 (dd, J = 8.8, 2.6 Hz, 1H), 7.32 (d, J = 2.6 Hz, 1H), 7.92 (d, J = 9.0 Hz, 1H), 10.2 (s, 1H).
