General procedure: To a suspension of methyl (Z)-1-acetyl-3-(ethoxy(phenyl)methylene)-2-oxoindoline-6-carboxylate (6) (500 mg,1.368 mmol) in DMF (3.5 mL) was added N-(4-aminophenyl)-N-methyl-2-(4-methylpiperazin-1-yl)acetamide (14) (395 mg,1.505 mmol, 1.1 equiv.) at RT. After heating the reaction mixture at 80 C for 1 h, it was allowed to cool to RT. Piperidine (297 lL,3.010 mmol, 2.2 equiv.) was then added and stirred for 2 h. Volatiles were removed in vacuo and water was added to the obtained residue and stirred for 15 min. The precipitate was then filtered under suction and cake was washed with water, then with minimum amount of cold methanol, and then ether. The obtained product was purified by column chromatography (neutral Al2O3,0-10% methanol in CH2Cl2) to afford 532 mg (72%) of target molecule 15., 262368-30-9

A heterogeneous mixture of methyl (E)-1-acetyl-3-(methoxy(phenyl)methylene)-2-oxoindoline-6-carboxylate (25 g ), N-(4-aminophenyl)-N-methyl-2-(4-methylpiperazin-1-yl)acetamide (22.4 g, for preparation, see e.g. US 6762180 Bl or US 8304541 B2), methanol (200 ml) and N,N-dimethylformamide (50 ml) was stirred and heated to reflux for 3-4 hours. A clear brown solution was obtained. A sample was drawn and analyzed for the presence of the limiting starting material (nmt 1 %). Piperidine (10.5 ml) was then added and the mixture was stirred under reflux for another 30-60 minutes. The product precipitated out during the stirring. The reaction mixture was analyzed for the intermediate and once nmt 1 % remained as determined by HPLC, the mixture was cooled to 0 C and stirred from 2 h to overnight. The solids were isolated by filtration and washed twice with methanol (75 ml per wash), then dried in a vacuum oven at 40 C overnight to obtain methyl (Z)-3-(((4-(N-methyl-2-(4-methylpiperazin-1-yl)acetamido)phenyl)amino)(phenyl)methylene)-2-oxoindoline-6-carboxylate (33.7 g, 88 %, 99.8 a-%) as a bright yellow solid., 262368-30-9

Preparation of the Final Compounds: (cr) 3-Z-[1-(4-(N-(4-methyl-piperazin-1-yl-methylcarbonyl)-N-methyl-amino)-anilino)-methylene]-6-ethylcarbamoyl-2-indolinone 160 mg 3-(1-hydroxy-methylene)-6-ethylcarbamoyl-2-indolinone (starting material VIII) and 543 mg N-[(4-methyl-piperazin-1-yl)-methylcarbonyl]-N-methyl-p-phenylendiamine are dissolved in 3 ml of tetrahydrofuran, 506 ml trimethylsilylimidazole are added and the mixture is stirred for 25 minutes at 170 C. in a microwave oven. After cooling the solvent is evaporated and the residue is purified over an aluminum oxide column (activity 2-3) with methylene chloride/ethanol (19:1) as eluant. The residue is recrystallized from ether and vacuum-dried at 80 C. Yield: 0.17 g (52% of theory), Rf value: 0.60 (aluminum oxide, methylene chloride/methanol=9:1) Melting point: 255-260 C.
