3-(2-ethyl-1,3-dioxolan-2-yl)propanal (1.5 eq) (prepared as described by Kuehne, M.E. et al J. Org. Chem. 1981, 46, 3443) was added to a stirred solution of l-(tert-butyl) 2-methyl piperazine- 1,2-dicarboxylate (1.0 eq) in MeOH (0.1 M), followed by AcOH (until pH = 6) and stirred for 1 h. Then NaBH3CN (1.5 eq) was added and the mixture was stirred at 20 C for 16 h. Two subsequent additions of 3-(2-ethyl-l,3-dioxolan-2-yl)propanal (0.5 eq + 0.5 eq) and NaBH3CN (1.0 eq + 1.0 eq) after 1 h and 2 h were done. MeOH was removed under reduced pressure. The residue was dissolved with DCM, washed with aq. sol. NaHC03, brine, dried and concentrated to give the title compound as yellow oil which was used in the next step without further purification.
