To a stirred solution of 1 -(tert-butyl) 3-methyl piperazine-1 ,3-dicarboxylate (CAS Number 129799-08-2; 0.500 g, 2.05 mmol) in DMF (10 ml) was added TEA (0.300 ml, 2.670 mmol) at 0C. Methanesulphonyl chloride (0.300 g, 2.67 mmol) was added dropwise to the reaction mixture at 0C, warmed to rt and stirred for 4 h. The resulting reaction mixture was poured into water (40 ml) and extracted with EtOAc (2 x 30 ml). The combined organic phase was collected, dried over Na2S04, filtered and concentrated under reduced pressure yielding 1 -(tert-butyl) 3-methyl 4- (methylsulfonyl)-piperazine-l ,3-dicarboxylate (0.120 g, 0.372 mmol).
