A new chem. library based on the hybridization of cholic acid with the heterocyclic moiety 1,3,4-oxadizoles I [R = 4-(phenyl)piperazin-1-yl, morpholino, benzyl(methyl)amino, etc.] were synthesized, and tested for antimicrobial activity against Gram-pos., Gram-neg. bacteria, and fungi. Among the synthesized compounds, the most potent derivatives against S. aureus were I [R = dibutylamino, 4-methylpiperazin-1-yl, pyrrolidin1-yl, 4-(2-fluorophenyl) piperazin-1-yl] with MIC values between 31 and 70μg/mL, while compound I [R = pyrrolidin1-yl] was the most active one against Bacillus subtilis with a MIC value of 70μg/mL. Interestingly, compounds I[R = 4-(phenyl)piperazin-1-yl, dihexylamino] exerted selective activity against Gram-pos. bacteria. The synthesized compounds I showed good activity against A. fumigatus and C. albicans and compound I [R = dicyclohexylamino] exhibited selective activity against fungi only.
