To a vial containing (R) -tert-butyl 2-isobutylpiperazine-1-carboxylate (17-a) (50.8 mg, 0.210 mmol, commerically available from Matrix Scientific, Cairo, Egypt) , were added compound 10-d (48.3 mg, 0.220 mmol) , DMF (2.0 mL) and DIEA (100 mul, 0.57 mmol) . The mixture was stirred at RT for 19 h. The mixture was then filtered and purified by preparative, reverse-phase HPLC (eluting with ACN/water with 0.1%TFA) to afford (R) -tert-butyl 4- (5-cyano-2- (1-methyl-1H-pyrazol-4-yl) pyrimidin-4-yl) -2-isobutylpiperazine-1-carboxylate (17-b) as the TFA salt.
