Novel anilide and benzylamide derivatives of arylpiperazinylalkanoic acids as 5-HT1A/5-HT7 receptor antagonists and phosphodiesterase 4/7 inhibitors with procognitive and antidepressant activity

A library of novel anilide and benzylamide derivatives of omega-(4-(2-methoxyphenyl)piperazin-1-yl)alkanoic acids as combined 5-HT1A/5-HT7 receptor ligands and phosphodiesterase PDE4B/PDE7A inhibitors was designed using a structure-based drug design approach. The in vitro studies of 33 newly synthesized compounds (7-39) allowed us to identify 22 as the most promising multifunctional 5-HT1A/5-HT7 receptor antagonist (5-HT1AKi= 8 nM, K-b = 0.04 nM; 5-HT7 K-1 = 451 nM, K-b = 460 nM) with PDE4B/PDE7A inhibitory activity (PDE4B IC50 = 80.4 mu M; PDE7A IC50 = 151.3 mu M). Compound 22 exerted a very good ability to passively penetrate through biological membranes and a high metabolic stability in vitro. Moreover, the pharmacological evaluation of 22 showed its procognitive and antidepressant properties in rat behavioral tests. Enhancing the permeance and antifouling properties of thin-film composite nanofiltration membranes modified with hydrophilic capsaicin-mimic moieties

Enhancing the water permeance and improving the antifouling performance of developing thin film composite (TFC) nanofiltration (NF) membranes remains a great challenge for practical applications. In this work, a novel TFC membrane was fabricated by introducing a new capsaicin derivative (propyl 2-(acrylamidomethyl)-3,4,5-trihydroxybenzoate, PAMTB) into a polyamide (PA) layer during interfacial polymerization for the NF process. The abundant hydroxyl groups in the PAMTB molecules can compete with the amino group in piperazine (PIP) to react with acyl chloride in trimesoyl chloride (TMC), facilitating the formation of a smooth and hydrophilic membrane surface with an enlarged pore size to promote water permeance. Compared to the control TFC membrane, the PAMTB-incorporated membrane exhibited increased water flux from 80 L m-2h-1 to 115 L m-2h-1 and a steady rejection of above 98.0% with 2.0 g L-1 Na2SO4 as the feed solution under a pressure of 5.0 bar. Different measurement methods demonstrated that the TFC-PAMTB membrane exhibited superior antifouling properties and long-term stability. In particular, the permeation flux recovery was approximately 100% after simple hydraulic washing with HA solution as the foulant. This work introduced a new approach to exploit capsaicin-mimic moieties in water treatment applications.

Diisocyanates are occupational contaminants and known sensitizers causing irritation (skin and respiratory tract) as well as occupational asthma. Because of their physicochemical properties (semi-volatile and high reactivity) and low occupational limits, diisocyanate exposure evaluation is still a challenge nowadays for industrial hygienists and laboratories. The objective of this study was to compare the methylene diphenyl diisocyanate (MDI) concentrations measured by five methods using different collection or derivatization approaches in an oriented-strand board (OSB) factory. The methods used were: OSHA 47 (filter, 1-(2-pyridyl)piperazine) (OSHA), Asset EZ4-NCO (denuder and filter, dibutylamine) (Asset), Iso-Chek (double-filter, 9-(N-methylaminomethyl) anthracene and 1,2-methoxyphenylpiperazine), DAN (filter, 1,8-diaminonaphthalene), and CIP10 (centrifugation, 1,2-methoxyphenylpiperazine). Real-time monitoring of particle concentration and size distribution was performed to explain the potential bias between methods. The comparison study was performed over 3 consecutive days, generating at least 18 replicates for each of the 5 methods. The results of each methods were compared using linear mixed effect modeling. Compared to Asset, which yielded the highest concentrations overall, the OSHA method provided the smallest bias with -18% (95% CI [-61;24]) (not significant) for MDI monomer and the DAN method provided the smallest bias with -30 (95% CI [-70;9]) (not significant) for Total Reactive Isocyanate Group (TRIG). The CIP10 and Iso-Chek methods provided the largest biases for MDI monomer (-83% (95% CI [-115;-51]) and -78% (95% CI [-110;-46]), respectively) as well as for TRIG (-87% (95% CI [-120;-55]) and -75% (95% CI [-107;-44]), respectively). The underestimations of the CIP10 and Iso-Chek were explained by its inefficient sampling principle for fines particles and the use of a non-impregnated filter to collect aerosol MDI, respectively. This study confirms that impregnated filter, including denuding device such as the Asset EZ4-NCO sampler, collects the MDI-coated wood particles and MDI vapor with similar efficiency. It also demonstrates for the first time in this type of MDI emission a significant agreement for TRIG concentration between the DAN method in the impregnated filter configuration and an international standard one such as Asset.

In vitro efficacy of anthelmintics on Angiostrongylus cantonensis L3 larvae

Angiostrongylus cantonensis is the leading cause of eosinophilic meningitis worldwide, with life-threatening complications if not managed correctly. Previous in vitro studies have utilized change in motility patterns of adult female worms to assess the efficacy of anthelmintics qualitatively. However, it is the third stage larvae (L3) that are infectious to humans. With differential staining using propidium iodide penetration as the indicator of death, we can distinguish between dead and live larvae. This assay has enabled us to quantify the in vitro efficacy of nine clinically established anthelmintics on A. cantonensis L3. All drugs were tested at a 1 mm concentration. Piperazine and niclosamide were ineffective in inducing larval death; however, albendazole sulfoxide, pyrantel pamoate, diethylcarbamazine, levamisole and praziquantel were effective as compared to unexposed controls (P < 0.05). Ivermectin and moxidectin did not induce significant levels of mortality, but they considerably reduced larval motility almost immediately. In vitro and in vivo antimicrobial activities of a novel piperazine-containing benzothiazinones candidate TZY-5-84 against Mycobacterium tuberculosis

The MIC value of TZY-5-84 against M. tuberculosis H37Rv ranged from 0.014 to 0.015 mg/L, lower than those of INH, RFP and BDQ. Five susceptible and thirteen drug-resistant clinical isolates were also susceptive to TZY-5-84. It had anti-tuberculosis activity against intracellular bacilli in infected macrophage model. It exhibited its activity in time-dependent manner and against intracellular bacilli in infected macrophage cells. However, the MIC of TZY-5-84 against three laboratory PBTZ169-induced resistant isolates increased four-fold increment compared to that of H37Rv. No antagonism was observed in any combination between TZY-5-84 and seven commonly used anti-tuberculosis drugs in an in vitro checkerboard assay. In murine infection model, TZY-5-84 at lower dosage (12.5 mg/kg) was found to be comparatively efficacious as PBTZ169 at 25 mg/kg. Our research suggests TZY-5-84 can be a promising preclinical candidate for further study on TB treatment.

Probing the charged nature and ion-exclusion mechanism of fluorine-enriched non-ionogenic polyamide derived thin film composite nanofiltration membranes

A thin film composite (TFC) nanofiltration (NF) membrane comprising a novel fluorine-enriched polyamide (FPA) as a skin layer polymer has been developed by in situ interfacial polycondensation between a cyclo-aliphatic diamine monomer, piperazine, and an acyclic fluorodiacyl chloride monomer, tetrafluorosuccinyl chloride. Instrumental investigations carried out by FTIR and XPS showed that the structure of FPA is devoid of intra- as well as inter-molecular H-bonds, which seems to be a rare finding in NF type membranes with decent performances. Stereoelectronic effects of the FPA structure, owing to the configurational mobility of polymer chains, electron-withdrawing ability of fluorine and lack of cohesive non-covalent type interactions, strongly influenced the surface morphology, topography and hydrophilicity of the TFC-NF membranes. The active skin layer of the membranes exhibited uniform charge-atmosphere in spite of not having any fixed charged groups in the polymer structure constituting it. Zeta potential measurement of the membranes revealed low values (-4.4 to -7 mV), indicating their low surface charge. However, a high Donnan-exclusion was observed towards a solute with multivalent anion (Na2SO4, solute rejection: 87-93%) as opposed to that with monovalent anion (NaCl, solute rejection: 23-26%), with a solvent flux as high as 54 L m(-2) h(-1) at 15 bar transmembrane pressure. The current class of TFC-NF membranes with unique physicochemical attributes thus provides a promising scope for a variety of nanofiltration applications.

Quantitative profiling of eicosanoids derived from n-6 and n-3 polyunsaturated fatty acids by twin derivatization strategy combined with LC-MS/MS in patients with type 2 diabetes mellitus

Eicosanoids derived from n-6 and n-3 polyunsaturated fatty acids (PUFAs), serving as important signaling molecules, are implicated in many physiological and pathological processes, including Type 2 diabetes mellitus (T2DM). However, the quantification of endogenous eicosanoids is challenged by high structural similarity, low abundance in biological sample and poor electrospray ionization efficiency. In the current study, a sensitive and accurate liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed to quantify 65 eicosanoids derived from n-6 and n-3 PUFAs in plasma samples using twin derivatization strategy with a pair of reagents, 5-(dimethylamino) naphthalene-1-sulfonyl piperazine (Dns-PP) and (diethylamino) naphthalene-1-sulfonyl piperazine (Dens-PP). Dns-PP-derivatized plasma sample was mixed with the equal volume of Dens-PP-derivatized eicosanoid internal standards for LC-MS/MS analysis in multiple reaction monitoring (MRM) mode. After Dns-PP derivatization, the ionization efficiency and separation performance were substantially improved, resulting in the enhanced sensitivity by 446- to 1009-folds compared to intact eicosanoids. The quantitative accuracy determined by twin derivatization method was found to be comparable with stable isotope labeled internal standards (SIL-IS) method. The newly proposed method was successfully employed to quantify the target eicosanoids in plasma samples from healthy controls and the patients with T2DM. N-6 PUFA-derived eicosanoids, PGF2 alpha, PGD2, PGE2, PGA2, PGB2, 20-HETE and LTC4, significantly increased in plasma sample of T2DM patients. Oppositely, n-3 PUFA-derived eicosanoids, RvE1, 12(S)-HEPE and RvD1, remarkably decreased. Spearman’s correlation analysis indicated the strong correlations between these highlighted eicosanoids and clinical parameters of T2DM. Collectively, the sensitive and reliable eicosanoid quantification method may facilitate to elucidate the characteristics of eicosanoid metabolism and understand the role of eicosanoids in the pathogenesis of T2DM and other diseases.
