To a solution of methyl 4-chloro-3 -((8-(4-methoxybenzyl)-7-oxo-7,8-dihydro-6H-pyrimido[5,4-b][1,4]oxazin-4-yl)oxy)benzoate (step 1 intermediate) (210 mg, 0.46 mmol)and 4-((4-ethylpiperazin- 1 -yl)methyl)-3 -(trifluoromethyl)aniline (Intermediate Cl) (110 mg, 0.38 mmol) in toluene (3.0 mL) was dropwise added trimethyl aluminium solution (2M in toluene, 768 jiL, 1.54 mmol) at RT. The mixture was stirred for 3-5 h at RT. The mixture was quenched with aqueous ammonium chloride solution and extracted twice in ethyl acetate. Thecombined organic layers were washed with water followed by brine. The solution was dried over anhydrous sodium sulfate, filtered and concentrated under reduced pressure.
